Waters ACQUITY UPLC System with the PDA Detector.

Page 2: ACQUITY UPLC PhoTodIode ArrAY deTeCTor Console Software ... · Console Software The ACQUITY UPLC PDA Detector is equipped with a customizable instrument console, for both Empower

optimized for UPLC separations

Waters ACQUITY UPLC Systems have been shown to significantly reduce run times. The ACQUITY UPLC PDA Detector, coupled with innovative ACQUITY UPLC column chemistries and separations modules, leverages ultra-low dwell volumes and low-noise, high-speed detection to bring you productivity, sensitivity, and resolution that are unprecedented in HPLC techniques today.

Additionally, the PDA Detector may be interfaced directly with mass spectrometers. The ACQUITY UPLC System’s low dispersion fluidics eliminate the need to remove the optical detector, or to split the flow path between the two detectors, in order to maintain optimal resolution.

Trace Impurity detection

The Waters ACQUITY UPLC PDA Detector has exceptional signal-to-noise ratios, high optical and digital resolution, and high-sensitivity library matching. This enables the ACQUITY UPLC System to map low levels of compounds and determine trace impurity levels within the compound’s peak. Unique Lamp Optimization Software optimizes light intensity and maintains minimal noise level across the entire deuterium spectrum over the lifetime of the lamp. The need to compromise optical bandpass and linear range simply to reduce noise is eliminated.

Superior linear range

The ACQUITY UPLC PDA detector can acquire chromatographic and spectral data at wavelengths 190 to 500 nm and up to 2.0 AU, without compromising linearity. Such conformity is crucial when comparing spectra across a broad wavelength and concentration range. Without it, disparate levels in common stability indicating methods cannot be quantified at the wavelength that offers the best signal-to-noise and minimal interference.

Independent optimization of data rate and Filtering Constants

UPLC Systems can generate peak widths as low as one second half-height, a detection challenge for which the ACQUITY UPLC PDA Detector is specifically designed. The detector has a high sampling rate to perform accurate and reproducible integration of the analyte peak, while allowing for independent optimization of the data rate and the filtering constants for the best sensitivity and resolution.

Light rays entering the liquid core of the flow cell are internally reflected when they meet the Teflon AF boundary. These rays are transmitted through the cell without loss, except for absorption by the sample.

Light-guiding flow cell technology

Small bore high-capacity ACQUITY UPLC Columns produce small volume peaks. To avoid band spreading and maintain concentration, the detector flow cell volume must be correspondingly low. To achieve the required volume reduction with conventional absorbance detector flow cells, the pathlength must be reduced, which, as predicted by Beer’s law, results in a loss of sensitivity.

Waters has specifically designed a low volume light-guiding flow cell for the ACQUITY UPLC PDA Detector that has optimum path length and high light throughput. The cell uses Teflon® AF, utilizing total internal reflection principles, to improve light transmission efficiency by eliminating internal absorption.

Plotting the detector response versus concentration demonstrates the linear range and linearity for high concentration actives and low concentration impurities.

Optimizing for sensitivity and resolution with different data rates and different filter constants provides the best results. The simultaneous collection of 3D data provides the spectral information needed for library matching and peak purity analysis.

The ACQUITY UPLC PDA detector allows quantitation of impurities at levels down to 0.004%.
